Final Separation & Quantification of Desired Compounds

  1. Methanol is added to the dried down sample extract and the tubes are sonicated to dissolve the samples.
  1. The dissolved samples are filtered and placed into vials.
  1. All vials are placed in a carousel so they can be measured one after the other.
  1. The carousel is put inside the controller where small volumes of each sample will be injected.
  1. Samples run through the column (on the left) and the UV absorbance of chemicals that are separated in the column is measured with the detector (bottom right)
  1. Absorbance is recorded on chromatograms and the compounds can be identified by the time they are recorded (31.85 minutes here for ajmalicine). The area under the peaks corresponds to the amount of compound present in a sample.
